If you are driving:
There are two US Highways leading in from the north, US Route 281 and US Route 77.
US Route 281 stretches from the North Dakota/Canadian border (Hwy 5 in Manitoba, Canada) to the Texas/Mexican border. This scenic route passes through N. Dakota, S. Dakota, Nebraska, Kansas, Oklahoma and Texas with its southernmost point being the Pharr-Reynosa International bridge in Pharr, Texas.
US Route 77 stretches from Sioux City, Iowa to the Veteran's International Bridge at the Mexican Border with Brownville, Texas; passing through Nebraska, Kansas, Oklahoma and Texas.
Both US Route 281 and US Route 77 intersect with all East-West Interstate Highways in the Midwestern US. So you can head either East or West, depending upon your starting point, connect with either 281 or 77 and head South until you are here.
There are of course ways to shorten the distance by traveling diagonal routes to reach either 281 or 77. For example, starting from Kansas City, MO take Interstate 35 southwest to San Antonio, Texas then connect with 281 south at which point you are about three hours away from the Valley.
Using a good old road map or web based map system will allow you to navigate the shortest or most interesting route to follow from wherever you are originating.
Flying in:
There are three airports in the Rio Grande Valley;
The McAllen-Miller International Airport in McAllen, Texas
Valley International Airport in Harlingen, Texas
Brownsville South Padre Island International Airport in Brownsville, Texas.
